ABSTRACT:
Respiratory disorders such as allergic rhinitis, sinusitis, and upper airway inflammation require therapeutic approaches that provide rapid onset of action and improved patient compliance. Herbal actives delivered through nasal in-situ gelling systems offer advantages such as prolonged residence time, sustained release, and enhanced mucosal absorption. Ocimum sanctum (Tulsi) possesses potent anti-inflammatory, antimicrobial, immunomodulatory, and antioxidant properties, making it a promising candidate for nasal formulations. The present study aimed to develop and characterize a thermosensitive nasal in-situ gel of Ocimum sanctum extract using Poloxamer 407 and HPMC as gelling and viscosity-modifying agents. A hydroalcoholic extraction method was optimized to obtain a phytochemically rich Tulsi extract. The formulated gels were evaluated for pH, clarity, viscosity, gelation temperature, drug content, in-vitro gelation, spreadability, rheology, in-vitro drug release, antimicrobial activity, and stability. Results indicated the successful development of a stable nasal in-situ gel with desirable physicochemical and release characteristics suitable for respiratory disorders.
